چکیده

The proto-oncogene tyrosine kinase c-ROS is an orphan receptor whose normal expression pattern is tightly spatio-temporally restricted during development. In glioma, c-ROS mRNA expression is frequently ectopically up-regulated. In this study, we determined by immunohistochemical means that c-ROS receptor protein is present in 25% of low-grade and 30% of malignant glioma tumor samples from tissue microarrays. We then explored the molecular basis for the up-regulation of c-ROS expression in these tumors. We identified and characterized the c-ROS gene promoter region and report that the ectopic expression of c-ROS in tumors is tied to hypomethylation of a CpG island in the c-ROS promoter. Bisulfite sequencing analysis in glioma tumor samples revealed that demethylation of the CpG island (-384 to -132 bp) correlated with c-ROS expression. Moreover, c-ROS expression could be activated by treatment of c-ROS-negative cells with the demethylating agent 5-aza-2'-deoxycytidine. These results establish a strong link between c-ROS promoter demethylation and gain of c-ROS expression and function in glioma. Our data suggest that epigenetic activation of c-ROS represents an important oncogenic mechanism for glioma initiation and progression and suggest that cautionary measures in the clinical use of 5-aza-dC for the treatment of glioma be taken into consideration. N-Acetylcysteine Compared to Metformin, Improves The Expression Profile of Growth Differentiation Factor-9 and Receptor Tyrosine Kinase c-Kit in The Oocytes of Patients with Polycystic Ovarian Syndrome

Objective Paracrine disruption of growth factors in women with polycystic ovarian syndrome results in production of low quality oocyte, especially following ovulation induction. The aim of this study was to investigate the effects of metformin (MET), N-acetylcysteine (NAC) and their combination on the hormonal levels and expression profile of GDF-9, BMP-15 and c-Kit, as hallmarks of oocyte qual...

Determination of CD117 Expression in Glial Tumors and Its Comparison between High Grade and Low Grade Tumors

Background and Objective: Gliomas are the most common primary brain tumors. Despite therapeutic advances, the majority of gliomas do not respond to either chemo or radiotherapy. CD117, the gene product of c-kit has been expressed in cells of glial tumors. Because gastrointestinal stromal tumors (GISTs) that express CD117 respond dramatically to treatment with tyrosine kinase inhibitors, ide...

Developmental expression of tyrosine kinase b in rat vestibular nuclear neurons responding to horizontal and vertical linear accelerations

Brain-derived neurotrophic factor (BDNF) is known to be crucial for the development of peripheral vestibular neurons. However, the maturation profile of the BDNF signal transducing receptor, tyrosine kinase B (TrkB) in functionally activated otolith-related vestibular nuclear neurons of postnatal rats remains unexplored. In the present study, conscious Sprague-Dawley rats (P4 to adult) were sub...

The Effect of Resistance Training Along with Royal Jelly Supplementation on Expression of Nerve Growth Factor and Tyrosine Kinase A Receptor in the Hippocampal Tissue of Alzheimer's Rats

Introduction: Current study aimed to investigate the effects of resistance training (RT) along with royal jelly (RJ) supplementation on hippocampal expression of nerve growth factor (proNGF) and p75 receptor in a rat’s model of Alzheimer's disease.  Method: 42 male Sprague-Dawley rats were treated with Trimethyltin chloride (8 mg/kg). Then, the rats were randomly divided into seven equal group...
